I would get a bike fitting from a professional fitter before anything else, measure my sitbones at home, and start searching for the perfect bike seat (saddle).

Minimum gear is a cycling specific helmet, gloves, and safety glasses. The glasses and gloves don't have to be cycling specific. They are to protect you in falls and from road debris. (You'll understand the first time a bug hits you in the face at 18 mph).
Also nice, good shorts and a wicking jersey. (TE is definitely the best place to find good fitting ladies cycling clothing). Plenty of stuff. But I don't believe you need anything special. Lycra shorts need to be snug to prevent chafing. Some ladies do like to wear a pair of loose capris or some such over the shorts when they first start. It's not necessary at all. But whatever makes you feel good.
A set of lights are a good idea. This is available on TE. Great deal.

Is the bike in good shape and well-maintained? Does it need a tune-up?
Check under bike maintenance for threads on how to wash, lube, and everything else your bike. I think you were actually referring to the wheel-set, not the tires. Check your tires and make sure they are sound, with no cuts or suspicious areas. And get a good, large floor pump. The small ones are harder to use. There are other threads for great stuff to buy to start out.
